The head of the lake is all Lake Chelan National Recreation Area (Lake Chelan National Recreation Area | National Park Foundation). I suspect you can ride all the road and former road in the Stehekin Valley, but I believe riding the trails would be poaching. I'm sure the residents of Stehekin ride bikes on the roads up there.
I don;t know about the rest of the gang here, but I have been using wilderness.net's map as a tool to see what can and can't be done. Just yesterday I was looking at Stehekin and I still don't see a way into it other than taking the boat. Is there even a dirt road that leads to Stehekin from the outside?
