not 100% as I don't play with Shimano stuff much anymore and don't play roadie at all... but A guy I ride with is using road cassette and derailleur with mtn (LX If I remember) shifters and it runs fine...
Going to try and bring Trail Tire TV back. go take a look...

10 speed or 9? 10 speed mtb doesn't work with road 10 speed.

    For Shimano
    A 9spd MTB RD will work with a 10spd road brifter (brake/shifter).

A road (Ultegra) RD should work with a 9 spd MTB shifter. ( 9 and 10spd road RD have the same pull ratio).

10spd MTB components work best when used together, no mix and match there are some exceptions.
Duct tape iz like teh Force. It has a Lite side and a Dark side and it holdz the Universe together.

Anything branded "dyna-sys" needs to be used with the same unit on the other end.

Shimano RD's are all the same cable pull except for the dyna-sys stuff. So you can cross road and mountain all the way back to 7 speed and stay compatible. However you can't run them with brifters, those need special RD's that are super hard to find. But if you want to use an integrated brake/shifter for a drop bar (normally people just call them SHIFTERS) it'll work just fine.

If you just want to use a road cassette and chain with MTB shifter and RD it will all be fine as long as all the bits are 10 speed (or 9 speed, or 8 speed). All cassettes with the same number of cogs (road or MTB) have the same spacing between cogs.

As others have said, Dynasys shifters and RDs work only with each other. Pretty well all other Shimano shifters & RDs will cooperate, apart from Dura-Ace 740x (6,7,8 speed).

If you will be using a particularly small cassette it my help shifting if you loosen the B-tension screw to bring the top pulley closer to the cogs.
